    Operant Sensation Seeking Requires Metabotropic Glutamate Receptor 5 (mGluR5)

    Pharmacological and genetic studies have suggested that the metabotropic glutamate receptor 5 (mGluR5) is critically involved in mediating the reinforcing effects of drugs of abuse, but not food. The purpose of this study was to use mGluR5 knockout (KO), heterozygous (Het), and wildtype (WT) mice to determine if mGluR5 modulates operant sensation seeking (OSS), an operant task that uses varied sensory stimuli as a reinforcer. We found that mGluR5 KO mice had significantly reduced OSS responding relative to WT mice, while Het mice displayed a paradoxical increase in OSS responding. Neither KO nor Het mice exhibited altered operant responding for food as a reinforcer. Further, we assessed mGluR5 KO, Het and WT mice across a battery of cocaine locomotor, place preference and anxiety related tests. Although KO mice showed expected differences in some locomotor and anxiety measures, Het mice either exhibited no phenotype or an intermediate one. In total, these data demonstrate a key role for mGluR5 in OSS, indicating an important role for this receptor in reinforcement-based behavior

    Generating dialogical practices in mental health : experiences from southern Norway, 1998-2008

The development of new strategies frequently involves challenges regarding procedures and treatment as well as a view of knowledge and humanity. Dialogical practices might provide a solution for these challenges not only because of its procedures but also due to its attitudes toward service users. The aim is to explore the implementation of three dialogical practice programs in Southern Norway from 1998 to 2008 and to critically analyze and discuss the authors' experiences during the implementation process. Three different programs of dialogical practices were initiated, established, and evaluated within the framework of participatory action research. Sustainable changes succeed individually and organizationally when all participants engage as partners during the implementation of new mental health practices. Generating dialogic practice requires shared understanding of the Open Dialogue Approach (ODA) and collaboration between professional networks and among the leaders. Developing a collaboration area that includes service users in all stages of the projects was one of the essential implementation factors. Other factors involved a common vision of ODA by the leaders and the actors, similar experiences, and a culture of collaboration. However, ODA challenged traditional medical therapy and encountered obstacles to collaboration. Perhaps the best way of surmounting those obstacles is to practice ODA itself during the implementation process

    QTLs for fire blight (Erwinia amylovora) resistance in Pyrus ussuriensis.

    Fire blight is the most devastating bacterial disease of apples and pears. The need for highly disease-resistant cultivars is more pressing than ever. Genetic disease immunity or resistance is recognized as an important feature of integrated pest management (IPM). In this study, we used an F1 progeny of Pyrus communis \u2018Doyenne du Comice\u2019 7 Pyrus ussuriensis to build parental genetic maps and to identify quantitative trait loci (QTLs) related to fire blight resistance. Linkage maps were constructed using a set of microsatellites and enriched with amplified fragment length polymorphism (AFLP) markers. For the first time QTLs for fire blight resistance in P. ussuriensis were identified by the Kruskal-Wallis test in its LGs U9, U_a, U_e, Ug, and U11. QTLs identified in LGs U_e and U_g are linked to AFLP-RGA markers that additionally confirms the presence of resistance genes in these LGs. Despite the high susceptibility of P. communis \u2018Doyenne du Comice\u2019, four QTLs for fire blight resistance in this cultivar were localized in LG K3, K4, K11, and K_a. These data confirm the existence of the interspecific transgression phenomenon, also previously observed on the basis of infection level evaluation of seedlings belonging to the studied hybrid family

    International audienceFull-length cDNA clones corresponding to the RNA1 and RNA2 of the Polish isolate MJ of Tomato black ring virus (TBRV, genus Nepovirus) were obtained using a direct recombination strategy in yeast, and their complete nucleotide sequences were established. RNA1 is 7358 nucleotides and RNA2 is 4633 nucleotides in length, excluding the poly(A) tails. Both RNAs contain a single open reading frame encoding polyproteins of 254thinspkDa and 149thinspkDa for RNA1 and RNA2 respectively. Putative cleavage sites were identified, and the relationships between TBRV and related nepoviruses were studied by sequence comparison
